Animals were immunized with whole bacterium antigen from E. coli O157:H7 (ATCC 43895).
Animals were boosted with partially purified H7 flagellin.
Spleen cells were fused with P3X63Ag8.653 mouse myeloma cells.
The antibody is specific for E. coli H7 antigen. It also reacts with flagellin from some E. coli H23 and H24 strains, which are antigenically closely related to the H7 antigen.
High titer, high affinity and large volume ascites can be produced in mice injected intraperitoneally with these hybridoma cells.
The antibody has been used in ELISA, Western Blot, lateral flow and chemiluminescense assays.
Bacterial flagellum expression or motility should be promoted prior to use of this monoclonal antibody to avoid false negative reactions due to lack of target antigen.
