Restriction digests of the clone give the following sizes (kb): EcoRI--5.45, 1.2; HindIII--5.0, 1.65; PstI--6.4, 0.25; BamHI--6.65.
EST01828 includes sequences that translate into homeodomains, with the most similarity to Drosophila otd. It also includes a splice acceptor sequence that begins where the sequence of EST1828 diverges from otd.
EST01828 matches PIR accession A35912 with 52.77% identity and 75% similarity over the sequence range: 242-135.
Isolated from a directional library constructed by B. Soares, Columbia University.
First strand synthesis was primed with an oligo(dT)-NotI primer. Much of the priming occurred by the NotI portion of the primer, so although the clones are directional, the NotI site is not necessarily the 3' end of the message.
